Abstract:

Background & Aims: In this study, using a liquid-liquid microextraction method for pre-concentration trace amounts of aflatoxins, the amount of Aflatoxins B1 and B2 in powdered milk was determined. Determination of aflatoxins was done by using high-performance liquid chromatography coupled with fluorescent detector. Method: Samples were extracted by immunoaffinity column (IAC) clean-up, and their eluents were used as dispersants of the subsequent DLLME, for further enrichment of aflatoxins. Various parameters (the type of elution solvent, the type and volume of extraction solvent and disperser solvent, extraction time and centrifugation time) that affect the efficiency of two steps were optimized. Results: Under the optimum conditions, the calibrations for B1 and B2 were found to be linear in the range of 0.03-5.0 and 0.006-1.0 ng ml-1 with 0.98 and 0.99 coefficient of estimation (R2), respectively. Conclusion: The results showed that dispersive liquid–liquid microextraction combined with HPLC is a selective, simple, sensitive and effective analytical method for the pre-concentration and determination of ultra trace amounts of aflatoxins. The method is suggested for pre-concentration and determination of B1 and B2 aflatoxins in milk powder

Determination of aflatoxins B1 and B2 using ion mobility spectrometry.

This paper describes total determination of aflatoxins B1 and B2 in pistachio samples using corona discharge ion mobility spectrometry (IMS). A new injection port was designed to introduce liquid samples into the IMS. Using this port, the liquid solvent (methanol) containing standard aflatoxins or an extracted sample was directly introduced into the injection port. Calibration curves resulting ...

Determination of Aflatoxin M1 in Pasteurized Liquid and Powdered Milk Products Imported to Iran

Mycotoxins are the secondary metabolites of molds and have adverse effects on humans, animals, and crops, resulting in illnesses and economic losses. Aflatoxin M1 (AFM1) is a hepatocarcinogen found in the milk from animals that have consumed feeds contaminated with aflatoxin B1 (AFB1). Milk is a highly nutritious food and is a source of necessary macro- and micro-nutrients for the growth, devel...

Determination of aflatoxin M1 levels in raw milk samples using ELISA and high-performance liquid chromatography in Qazvin, Iran

Use of raw milk and dairy products containing aflatoxin M1 (AFM1) has led to concern inconsumers. The present study determined the AFM1 in raw cow's milk in Qazvinprovince. In this research, 170 raw cow's milk were collected from dairy farm, dairyfactories, milk collection centers, and milk supply centers in Qazvin province during coldseasons in 2013, and all samples were examined for AFM1. The...

Biosynthetic relationship among aflatoxins B1, B2, M1, and M2.

Aflatoxins are a family of toxic, acetate-derived decaketides that arise biosynthetically through polyhydroxyanthraquinone intermediates. Most studies have assumed that aflatoxin B1 is the biosynthetic precursor of the other aflatoxins. We used a strain of Aspergillus flavus which accumulates aflatoxin B2 to investigate the later stages of aflatoxin biosynthesis. This strain produced aflatoxins...
